Abstract: The application of bacteriophages is a promising alternative to conventional antibiotics. The strictly virulent and non-transducing bacteriophages are powerful tools to specifically treat pathogenic bacteria in food and agriculture. Taking into account the great diversity of bacteriophages, the isolation of a suitable biocontrol phage can be a challenging and time consuming task. In addition not every phage isolate can be used for biocontrol until it is not sufficiently characterized and approved. To enhance directed phage finding we developed a PCR driven approach to rapidly detect phages for biocontrol of Salmonella, Erwinia amylovora and other members of the Enterobacteriaceae. Isolated bacteriophages were initially characterized in terms of host range and genome size. Genomes of selected isolates were sequenced and subjected to in silico analyses. Because phage resistance can be observed after application, a combination of different phages, i.e. phage cocktails, with different host receptor specificities may be utilized. To identify phage receptors in target bacteria we established an easy to handle high throughput assay. Receptors of virulent bacteriophages with overlapping host ranges were identified and the effectiveness of the different bacteriophages, either applied alone, in different combinations, or after genetic modification, was tested in vitro, in food and in planta, depending on the targeted bacteria. It seems that Enterobacteria can be more efficiently controlled by phage cocktails than by single phage isolates.
