Ene reductases enable the asymmetric hydrogenation of activated alkenes allowing the manufacture of valuable chiral products. The enzymes complement existing metal- and organocatalytic approaches for the stereoselective reduction of activated C=C double bonds, and efforts to expand the biocatalytic toolbox with additional ene reductases are of high academic and industrial interest. Here, we present the characterization of a novel ene reductase from Paenibacillus polymyxa, named Ppo-Er1, belonging to the recently identified subgroup III of the old yellow enzyme family. The determination of substrate scope, solvent stability, temperature, and pH range of Ppo-Er1 is one of the first examples of a detailed biophysical characterization of a subgroup III enzyme. While sophisticated small-scale cultivation systems came to the market in the last few years, shake flasks maintain the economic alternative widely used in industry and academia. Yet, these vessels lack the sensors like turbidity, oxygen concentration and pH-values. Here we show how to overcome these hurdles. The first part covers measurement improvements for shake flasks with focus on back-scattered light for biomass growth monitoring and the utilization of sensor spots for a deeper insight into the “black box” shake flasks. We used plant cells as their growth often can’t be accurately monitored by simple offline OD measurements. The sensor spot measurements are further used to detect process events, enabling the prediction of metabolite depletion. In the second part, soft sensors based on engineering and biological characterizations of these vessels are introduced and explained, enabling researchers to predict process values and estimate growth curves.

Identification of coherent generators and the determination of the stability system condition in large interconnected power system is one of the key steps to carry out different control system strategies to avoid a partial or complete blackout of a power system. However, the oscillatory trends, the larger amount data available and the non-linear dynamic behaviour of the frequency measurements often mislead the appropriate knowledge of the actual coherent groups, making wide-area coherency monitoring a challenging task. This paper presents a novel online unsupervised data mining technique to identify coherent groups, to detect the power system disturbance event and determine status stability condition of the system. In this paper, symmetrical electric vehicle charging impacts in existing low-voltage distribution grid are investigated throughout proposed methodology and their results analysed. Symmetrical loading- and voltage-related impacts are assessed for the extensive grid. A synthetic EV mix pattern was used with the purpose to demonstrate a universal observation of charging impacts. These patterns were allocated quasi-randomly to the points of common coupling within the grid based on predefined scenarios - 8, 10, 12 and 20 percent. Subsequently, quasi-static time series simulations for a duration of one year in 10-minute time steps were executed. Consequently, this paper yields results, which offer practical insight in the maximum share of electric vehicle charging in low-voltage distribution grids and provide guidance for future decision-making of distribution grid operators.
Purpose – Voicing concerns and suggestions is crucial for preventing medical errors and improving patient safety. Research suggests that hierarchy in health-care teams impair open communication. Hierarchy, however, can vary with changing team composition, particularly during acute care situations where more senior persons join the team later on. The purpose of this study is to investigate how changes in hierarchy and leadership were associated with nurses’ voice frequency and nurses’ time to voice during simulated acute care situations. Design/methodology/approach – This study’s sample consisted of 78 health-care providers (i.e. nurses, residents and consultants) who worked in 39 teams performing complex clinical scenarios in the context of interprofessional, simulation-based team training. This paper presents recommendations for avoiding or eliminating unnecessary barriers to mobile application usage by older generations. The aim of this paper is to help ensure that the design of mobile applications is accessible for elderly persons. The recommendations are written in such a way as to be easily understandable for both app developers and app commissioners. In order to examine the needs of the 65-plus generation with regard to user-friendly applications, a research project was conducted with 10 elderly users. This consisted of three phases: a focus group, usability tests of four commonly-used mobile apps, and, finally, an open discussion with the test group about their experience. The objective of the research was to discover the views of older users and to identify specific barriers encountered when using mobile applications. Based on the results, ten key areas were identified for age-appropriate mobile app design.
Nachdem innerhalb der Diskursforschung auch die Diskurslinguistik zu einer eigenständigen Teildisziplin geworden ist, steht die Frage an, welches deren Beitrag für die Lösung gesellschaftlicher Problemstellungen sein kann. Aus diesem Grund schlagen wir ein transdisziplinäres Forschungsdesign vor, das theoretische, methodische und forschungspraktische Fragen gleichermaßen berücksichtigt: Es umfasst vier Forschungsmodule, deren korpuszentrierte Umsetzung und szenarienbasierte Nutzung wir exemplarisch vorstellen. Die transdisziplinäre Ausrichtung dieses Forschungsdesigns bringt dabei eigene Herausforderungen in der Forschungskommunikation mit sich.
